Purification of Human Serum Albumin by High Pressure Liquid Chromatography


Abstract
Albumin is purified from human serum or bovine serum by high pressure liquid chromatographic procedure in a yield of about 85% and a purity of up to 99%.
Human blood serum is treated with trichloroacetic acid-methanol solution. From the supernatant, by neutralization, the albumin is precipitated in a form undenatured and free from globulins. The precipitate is dissolved into phosphate buffer (pH 7.3) 10~20% w/v protein and applied to high pressure liquid chromatography equipped with single protein column I -125. Albumin fraction is collected and concentrated by ultra filtration. This combination method of high pressure liquid chromatography with protein column and ultra filtration provides an interesting alternative way to the purification of human serum albumin.
